Low expression of miR 126 is a prognostic marker for metastatic clear cell renal cell carcinoma. miR-126 is differentially expressed in many cancers including RCC and is down-regulated in metastatic versus primary ccRCC. We assessed the prognostic significance of miR-126 in 264 primary ccRCCs. We validated our results on an independent set of 481 ccRCCs miR-126 was down-regulated in metastatic versus primary tumors and in tumors of higher stage P = 0.005 or higher grade P = 0.002 miR-126 up-regulation was associated with significantly prolonged disease-free survival P < 0.001 and overall survival P = 0.015. For larger tumors >4 cm patients with higher miR-126 expression had significantly longer survival. The ccRCCs exhibited the highest miR-126 expression and papillary RCCs exhibited the lowest expression. We identified a number of miR-126 targets and pathways that are involved in carcinogenesis including the apoptosis signaling pathway. miR-126 is a promising prognostic marker in ccRCC that can distinguish between clear cell and papillary subtypes. Decreased expression of miR 126 correlates with metastatic recurrence of hepatocellular carcinoma. Based on microarray data we previously found that miR-126 expression was lower in tumor tissues of patients with post-LT HCC recurrence compared with non-recurrence. In this study we examined the expression of miR-126 in HCC samples from 68 patients who had undergone LT using quantitative real-time PCR and analyzed its correlation with clinicopathological features and prognosis of patients. Furthermore we performed experimental analyses to explore the involvement of miR-126 in HCC metastasis. We found that miR-126 levels were lower in tumor tissues of patients with post-LT HCC recurrence in comparison to patients with no-recurrence p = 0.009. Lower expression of miR-126 in HCC was associated significantly with tumor recurrence p = 0.011 and poor survival p = 0.009 of patients. Our study revealed that down-regulation of miR-126 plays an important role in HCC metastasis and suggest a potential application of miR-126 in prognosis prediction and HCC treatment

MiR-126-3p has been reported to be associated with poor prognosis in HCC. However the underlying mechanism of miR-126-3p in HCC remains unclear. The expression levels of miR-126-3p in HCC tissues and cells were detected by RT-PCR. Dual- luciferase reporter assay was conducted to confirm the direct binding of miR-126-3p and target genes. The changes of biomarker protein levels were examined by western blot and Immunohistochemistry. We observed that the miR-126-3p expression levels in HCC tissues and cells were significantly down-regulated. Furthermore overexpression of miR-126-3p significantly reduced the volume of tumor and microvessel density in vivo. Down regulation of microRNA 126 and microRNA 133b acts as novel predictor biomarkers in progression and metastasis of non small cell lung cancer. The aim of this study was to investigate the expression level of microRNA-126 miR-126 and microRNA-133b miR-133b and also their association with clinicopathological features in patients with non small cell lung cancer NSCLC. Total RNA was purified from NSCLC tissues and adjacent non-tumor tissues and then quantitative real-time PCR qRT-PCR was used to evaluate the expression rate of microRNAs. Furthermore the association of miR-126 and miR-133b level with clinicopathological features and prognosis were evaluated. Our findings showed that expression of miR-126 was decreased in NSCLC tissues compared with adjacent non-tumor tissues. In term of miR-126 our results showed that miR-126 was associated with tumor stage and lymph nodes metastasis P<0.05. Kaplan-Meier analysis and log-rank test indicated that patients with low expression of miR-126 and miR-133b had a shorter overall survival log-rank test; P<0.05. Multivariate Cox proportional hazards model revealed that low expression of miR-126 and miR-133b advanced tumor stage and lymph nodes metastasis were independent prognostic factors for overall survival of NSCLC patients. These findings suggested that miR-126 and miR-133b might play a key role in the progression and metastasis of NSCLC and would be applied as a novel therapeutic agent

Association of microRNA 126 expression with clinicopathological features and the risk of biochemical recurrence in prostate cancer patients undergoing radical prostatectomy. Numerous studies have suggested that microRNA-126 miR-126 is involved in development of various cancer types as well as in malignant proliferation and invasion. The aim of this study was to investigate miR-126 expression in PCa and its prognostic value for PCa patients undergoing radical prostatectomy. A series of 128 cases with PCa were evaluated for the expression levels of miR-126 by quantitative reverse-transcription PCR qRT-PCR. Kaplan-Meier analysis and Cox proportional hazards regression models were used to investigate the correlation between miR-126 expression and prognosis of PCa patients. Compared with non-cancerous prostate tissues the expression level of miR-126 was significantly decreased in PCa tissues PCa vs non-cancerous prostate: 1.05 ± 0.63 vs 2.92 ± 0.98 P < 0.001. Additionally the loss of miR-126 expression was dramatically associated with aggressive clinical pathological features including advanced pathological stage P = 0.001 positive lymph node metastasis P = 0.006 high preoperative PSA P = 0.003 and positive angiolymphatic invasion P = 0.001. Moreover Kaplan-Meier survival analysis showed that PCa patients with low miR-126 expression have shorter biochemical recurrence BCR-free survival than those with high miR-126 expression. Furthermore multivariate analysis indicated that miR-126 expression was an independent prognostic factor for BCR-free survival after radical prostatectomy. These findings suggest for the first time that the loss of miR-126 expression may play a positive role in the malignant progression of PCa. More importantly the downregulation of miR-126 may serve as an independent predictor of BCR-free survival in patients with PCa

We performed a miRNA microarray analysis by detecting six paired KS and matched adjacent healthy tissues using the 7th generation of miRCURYTM LNA Array v.18.0 Exiqon containing 3100 capture probes. We selected 10 significant differentially expressed miRNAs which were confirmed by quantitative real-time reverse transcription polymerase chain reaction qRT-PCR in 18 paired KS and matched adjacent healthy tissue specimens. Among the 3100 human miRNA probes in the microarrays we identified 170 differentially expressed miRNAs 69 upregulated and 101 downregulated miRNAs in KS versus adjacent healthy tissues. The objectives of this study are to investigate the expression of miR-126 and evaluate its effect on proliferation in undifferentiated thyroid carcinoma. Following transfection proliferation effects were observed through MTS proliferation assay and colony formation abilities.
