Many human malignant tumors demonstrate a low expression of miR-133b as noted in colorectal lung esophagus and bladder cancers but the role of miR-133b in bladder cancer is unknown. The expression of miR-133b in clinical bladder cancer specimens and adjacent normal tissues was confirmed by stem-loop RT-PCR. We also analyzed the relationship between miR-133b expression and clinicopathological factors of bladder cancer. The effect of miR-133b on T24 cell proliferation and apoptosis was measured by CCK-8 tests and flow cytometry respectively. The expression of miR-133b in bladder cancer tissues from 41 patients was significantly down-regulated P < 0.01; low expression of miR-133b was strongly associated with high-grade bladder cancer P < 0.01. We report that elevated microRNA-133b miR-133b acts as an oncogene in human cervical carcinoma to promote tumorigenesis and metastasis. The data indicates that upregulation of miR-133b shortens the latency of cervical carcinoma. TAp63 expression is downregulated in colon cancer and microRNA-133b is a transcriptional target of TAp63. Moreover TAp63 inhibits cell migration and invasion through microRNA-133b. TAp63 and microRNA-133b were able to suppress the metastasis of colon cancer. Combined downregulation of microRNA 133a and microRNA 133b predicts chemosensitivity of patients with esophageal squamous cell carcinoma undergoing paclitaxel based chemotherapy. microRNA-133a miR-133a and miR-133b located on chromosome 18 in the same bicistronic unit have been commonly identified as being downregulated in esophageal squamous cell carcinoma ESCC. miR-133a expression and miR-133b expression were examined in 100 newly diagnosed ESCC patients prior to treatment by quantitative real-time PCR. Expression levels of miR-133a and miR-133b were both significantly lower in ESCC tissues compared to adjacent noncancerous tissues both P < 0.001. Our data offer the convincing evidence that combined expression of miR-133a and miR-133b may predict chemosensitivity of patients with ESCC undergoing paclitaxel-based chemotherapy implying its importance in applying 'personalized cancer medicine' in the clinical treatment of ESCC. We also identified combined expression of miR-133a and miR-133b as an effective prognostic marker of this malignancy

Here we examine the role of miR-133b in gastric cancer. Quantitative real-time PCR analysis was performed in 140 patient gastric cancer tissues and 8 gastric cancer cell lines. In vivo effects of miR-133b were examined in an intraperitoneal mouse tumor model. Targets of miR-133b were predicted by bioinformatics tools and validated by luciferase reporter analyses western blot and quantitative real-time PCR. MiR-133b was significantly downregulated in 70% 98/140 of gastric cancer patients. Expression of miR-133b was negatively correlated with lymph node metastasis of gastric cancer in patients. MiR-133b is frequently decreased in gastric cancer. These results suggested that miR-133b plays an important role in gastric cancer metastasis

In this study we aimed to investigate the expression of miR-133b in a large number of GC samples and elucidate its role in GC carcinogenesis and the detailed mechanism. We used Taqman probe stem-loop real-time PCR to accurately measure the levels of miR-133b in 100 pairs of gastric cancer tissues and the adjacent non-neoplastic tissues. The target of miR-133b was identified by luciferase reporter assay and western blot. miR-133b was significantly down-regulated in GC cell lines and in GC tissues compared with adjacent normal tissues. Moreover lower-level of miR-133b was also associated with venous invasion and a more aggressive tumor phenotype. Our findings demonstrated that miR-133b was significantly down-regulated in GC tissues and exerted its tumor suppressor role in GC cells. Thus the differential expression of a panel of miRNAs in two pairs of gastric cancer tissues and their matched adjacent healthy tissues was investigated by performing a microarray analysis. To verify the results of this screen 56 gastric cancer tissues were analyzed for the selected miRNAs using reverse transcription-quantitative polymerase chain reaction RT-qPCR. A total of 31 differentially expressed miRNAs were identified in the miRNA array. Using RT-qPCR to verify these results it was determined that 10 miRNAs exhibited high mRNA expression levels and 13 miRNAs exhibited a low expression in the gastric cancer tissue samples while 8 miRNAs did not demonstrate an association with gastric cancer. Thus the microarray and RT-qPCR results demonstrated 74.2% 23/31 miRNAs agreement. It was identified that the expression levels of miR-551b-3p miR-133b miR-100-5p and miR-363-3p were significantly downregulated in the gastric cancer tissues and this downregulation was closely correlated with the degree of differentiation i.e tumor grade TNM stage and lymph-node metastasis P<0.05. In particular miR-551b-3p miR-133b miR-100-5p and miR-363-3p may act as tumor suppressors in the development of gastric cancer

Down regulation of microRNA 126 and microRNA 133b acts as novel predictor biomarkers in progression and metastasis of non small cell lung cancer. The aim of this study was to investigate the expression level of microRNA-126 miR-126 and microRNA-133b miR-133b and also their association with clinicopathological features in patients with non small cell lung cancer NSCLC. Total RNA was purified from NSCLC tissues and adjacent non-tumor tissues and then quantitative real-time PCR qRT-PCR was used to evaluate the expression rate of microRNAs. Furthermore the association of miR-126 and miR-133b level with clinicopathological features and prognosis were evaluated. On the other hand a lower expression of miR-133b was seen in NSCLC tissues when compared with adjacent non-tumor tissues. In term of miR-133b our finding indicated that decreased expression of miR-133b was correlated with advanced tumor stage and lymph nodes metastasis P<0.05. Kaplan-Meier analysis and log-rank test indicated that patients with low expression of miR-126 and miR-133b had a shorter overall survival log-rank test; P<0.05. Multivariate Cox proportional hazards model revealed that low expression of miR-126 and miR-133b advanced tumor stage and lymph nodes metastasis were independent prognostic factors for overall survival of NSCLC patients. These findings suggested that miR-126 and miR-133b might play a key role in the progression and metastasis of NSCLC and would be applied as a novel therapeutic agent

We examined the levels of miR-133b expression in ovarian carcinoma tissues and the human ovarian carcinoma cell lines A2780 A2780/DDP and A2780/Taxol respectively. The dual-luciferase reporter assay was used to detect the promoter activity of GST-π in the presence and absence of miR-133b. Dual-luciferase reporter assay showed that miR-133b interacted with the 3'-untranslated region of GST-π. Compared to controls the mRNA and protein levels of MDR1 and GST-π were downregulated after miR-133b transfection and upregulated after anti-miR-133b transfection. MicroRNA-133b may reduce ovarian cancer drug resistance by silencing the expression of the drug-resistance-related proteins GST-π and MDR1. In future the combination of miR-133b with chemotherapy agents may prevent the development of drug resistance in ovarian cancers

miR 1 and miR 133b are differentially expressed in patients with recurrent prostate cancer. First of all 20 recurrent and 20 non-recurrent PCa samples were profiled using miRNA microarray chips. Of the differentially expressed miRNAs miR-1 miR-133b and miR-145* were selected for further validation with qRT-PCR in a different set of 21 recurrent and 21 non-recurrent PCa samples. Our results demonstrated that miR-1 and mir-133b have been significantly downregulated in recurrent PCa specimens in comparison to non-recurrent PCa samples and have sufficient power to distinguish recurrent specimens from non-recurrent ones on their own. Here we report that the relative expression of miR-1 and mir-133b have been significantly reduced in recurrent PCa specimens in comparison to non-recurrent PCa samples which can serve as novel biomarkers for prediction of PCa progression
