Ectopic expression studies were carried out in MCF7 cells and miRNA expression evaluated in two additional cohorts of patients with breast cancer by in situ hybridization on tissue microarrays. CIA identified miR-187 as a key miRNA associated with poor outcome in breast cancer. In a test cohort n = 117 high expression of miR-187 was associated with a trend toward reduced breast cancer-specific survival BCSS; P = 0.058 and a significant association with reduced BCSS in lymph node-positive patients P = 0.036. In a validation cohort n = 470 high miR-187 was significantly associated with reduced BCSS in the entire cohort P = 0.021 and in lymph node-positive patients P = 0.012. Multivariate Cox regression analysis revealed that miR-187 is an independent prognostic factor in both cohorts cohort 1: HR 7.37; 95% confidence interval CI 2.05-26.51; P = 0.002; cohort 2: HR 2.80; 95% CI 1.52-5.16; P = 0.001 and in lymph node-positive patients in both cohorts cohort 1: HR 13.74; 95% CI 2.62-72.03; P = 0.002; cohort 2: HR 2.77; 95% CI 1.32-5.81; P = 0.007. miR-187 expression in breast cancer leads to a more aggressive invasive phenotype and acts as an independent predictor of outcome

MicroRNA 187 a downstream effector of TGFβ pathway suppresses Smad mediated epithelial mesenchymal transition in colorectal cancer. In this study expression of miR-187 was downregulated in colorectal cancer CRC compared with adjacent normal tissues. miR-187 could suppress the formation of aggressive phenotype in CRC and inactivate Smad pathway thus preventing EMT. TGFβ stimulation significantly suppressed the expression of miR-187 and overexpressed miR-187 counteracted the influence of TGFβ on cell phenotype and downstream pathway

However the underlying functions of microRNA-187 miR-187 in colorectal cancer CRC remain largely unexplored. Here we demonstrated that miR-187 was significantly down-regulated in CRC tissues and cell lines compared to their normal counterparts. By Kaplan-Meier analysis we revealed that decreased miR-187 expression was closely associated with shorter overall survival and relapse-free survival of patients with CRC. By gain- and loss-of-function studies we showed that miR-187 remarkably suppressed CRC cell proliferation migration invasion and promoted cell apoptosis

Based on the preliminary deep sequencing data we hypothesized that miR-187 may play an important role in ccRCC development. In this study we found that miR-187 was down-regulated in both tumor tissue and plasma of ccRCC patients. Lower miR-187 expression levels were associated with higher tumor grade and stage. All patients with high miR-187 expression survived 5years while with low miR-187 expression only 42% survived. Our data suggest that miR-187 is emerging as a novel player in the disease state of ccRCC miR-187 plays a tumor suppressor role in ccRCC

Two lung development related microRNAs miR 134 and miR 187 are differentially expressed in lung tumors. As some of the development-specific gene expression patterns could be observed in cancers we speculated that the expression pattern of lung development-specific miRNAs miR-134 and miR-187 might be altered in lung tumor samples. We employed a real-time RT-PCR approach to evaluate the expression alteration of two lung development-related miRNAs in lung tumor tissues. The effect of miR-187 overexpression on a lung carcinoma cell cycle was assessed using flow cytometry analysis. In contrast miR-187 showed a significant down-regulation P=0.008 in lung tumors. Altogether our data demonstrated an altered expression of two development-related miRNAs namely miR-134 and miR-187 in lung tumors for the first time. Moreover we have shown that miR-134 and miR-187 expression alternation were in accordance with their approved regulatory roles therefore these miRNAs could serve as new biomarkers with potential usefulness in lung cancer diagnosis and treatments
