In this study we show that miRNA-23b miR-23b acts as a tumor suppressor in bladder cancer. Quantitative real-time PCR analysis showed that miR-23b is significantly down-regulated in bladder cancer cell lines and tumor tissues compared to non-malignant cells and normal tissue samples. We also demonstrate that miR-23b expression has a potential to be diagnostic and prognostic biomarker in bladder cancer. High miR-23b expression is positively correlated with higher overall survival of bladder cancer patients as revealed by Kaplan-Meier analysis. ROC analysis showed that miR-23b expression can distinguish between normal and bladder cancer tissues. Further we elucidated the biological significance of miR-23b in bladder cancer. These results show that loss of miR-23b confers a proliferative advantage and promotes bladder cancer cell migration and invasion. Furthermore re-expression of miR-23b may be a beneficial therapeutic strategy for the treatment of human bladder cancer

Among methods for profiling levels of miRNAs next-generation sequencing NGS has an effective one for genome-wide profiles which not only can accurately quantify known miRNAs expression but also discovery novel miRNAs. In this paper we investigated that whether specific miRNAs were co-expressed in the serum and tissue of breast cancer BC patients as novel biomarkers by SOLiD sequencing. Different miRNA expression profiles of serum and tissue in breast cancer patients and control subjects were obtained by NGS -SOLiD sequencing. Real-time PCR was used to selected and validated candidate miRNA-biomarkers. Novel miRNAs were predicted by computational pipeline and validated by Northern blot analysis. Of genome-wide miRNA analysis using SOLiD sequencing 7 miRNAs were found to be co-upregulated i.e miR-103 miR-23a miR-29a miR-222 miR-23b miR-24 and miR-25 miR-222 was significantly increased in the serum of BC patients by further validationP<0.05 which may be a useful biomarker for differentiating BC patients from controls with receiver operating characteristic ROC curve area 0.67 of 95% CI=0.5649 to 0.7775. And next-generation sequencing will provide a robust platform for miRNA profilings

Up Regulation of Plasma miR 23b is Associated with Poor Prognosis of Gastric Cancer. This study was designed to evaluate the expression level of plasma miR-23b in patients with GC and investigate the relationship between plasma miR-23b expression level and the prognosis of GC. Quantitative real-time PCR qRT-PCR was performed to evaluate the expression level of plasma miR-23b in all participants. The association between miR-23b expression and clinicopathological factors as well as survival rates was analyzed. Receiver operator curve ROC analysis was carried out to evaluate the diagnostic performance of plasma miR-23b for GC.Univariate and multivariate Cox regression analyses were conducted to determine whether plasma miR-23b was an independent predictor of survival. The expression levels of miR-23b were upregulated in plasma samples from GC patients P<0.01 and were significantly associated with T stage distant metastasis and differentiation. Significantly shorter 5-year overall survival OS and disease-free survival DFS were observed in patients with higher expression of the miR-23b P<0.01. The area under the curve AUC of high expression of plasma miR-23b to diagnose GC was 0.80 95% CI: 0.74-0.86 P<0.001. Multivariate analysis revealed that enhanced expression of plasma miR-23b was an independent predictor of OS P=0.015 and DFS P=0.004. Our results indicated that plasma miR-23b was overexpressed in GC patients and high plasma miR-23b expression was associated with poor clinical outcome. Thus plasma miR-23b may serve as a potential diagnostic biomarker and therapeutic target for GC

Recently miR-23b has emerged as a promising new cancer biomarker but its role in lung cancer has not been established yet. The aim of this study was to determine the role of miR-23b in non-small cell lung cancer NSCLC and its relationship to the patient outcome together with MDR transporter proteins. The prognostic significance of miR-23b and MDR proteins was tested by Kaplan-Meier and Cox-regression analysis. Our results showed that miR-23b is mostly downregulated in NSCLC samples 57/62 and that its upregulation in tumors is connected with longer progression-free survival PFS; P = 0.065 and overall survival OS; P = 0.048. The Cox proportional hazard model revealed that the risk of death or relapse in NSCLC patients with miR-23b downregulation increases together with LRP/MVP expression and both risks decrease with miR-23b upregulation HRPFS = 4.342 PPFS = 0.022; HROS = 4.408 POS = 0.015. Our findings indicate that miR-23b especially in combination with LRP/MVP expression might serve as a suitable prognostic biomarker for NSCLC patients
