The differentially expressed microRNAs miRNAs miR were selected after hybridizing with miRNA microarray real time polymerase chain reaction real-time PCR was subsequently applied to confirm the expression of the selected miRs and PCR products were further cloned and sequenced to check their specificity. Real time PCR verified that the expression level of miR-10a and miR-220c in the CD34+ cells from the bone marrow of AML patients was 19.6% and 19.0% of that of CD34+ cells from mobilized peripheral blood of the normal control subjects. A variety of differentially expressed-miRNAs are existed between AML and normal control subjects CD34+ cells the expression of miR-10a and miR-220c was significantly down-regulated in the CD34+ cells from the bone marrow of AML patients

Upregulation of miR 20a and miR 10a expression levels act as potential biomarkers of aggressive progression and poor prognosis in cervical cancer. In the current study the expressions of miR-20a and miR-10a were evaluated in the cervical cancer tissues and in the corresponding noncancer tissues using qRT-PCR assay. Survival and Cox proportional hazards analyses were performed to find the association of miR-20a and miR-10a levels with prognosis in the patients. We found that miR-10a was overexpressed in cervical cancer tissues than those corresponding adjacent cervical tissues P < 0.05. Our findings showed that elevated expression of miR-10 was remarkably correlated with advanced FIGO stage P < 0.001 advanced histological grade 0.031 and increased size of the tumors P = 0.024. Kaplan-Meier analysis and log-rank test revealed that cervical cancer patients with high expression of miR-10 have shorter overall survival than patients with low expression log-rank test P < 0.001. Our findings showed that upregulation of miR-20a and miR-10a were correlated with aggressive progression and poor prognosis of cervical cancer

miRNA expression profile in primary gastric cancers and paired lymph node metastases indicates that miR 10a plays a role in metastasis from primary gastric cancer to lymph nodes. A miRNA array was used to compare the expression of miRNAs in primary gastric cancer and paired lymph node metastases. Moreover the target genes for miR-10a were predicted using bioinformatic methods. Based on the results four differentially expressed miRNAs were detected by the miRNA array. Compared with primary gastric cancer lymph node metastases displayed downregulated expression of miR-24-1* miR-510 and miR-1284 while the expression of miR-10a was upregulated. Consequently analysis found that the expression of miR-10a was associated with lymph node metastasis P=0.047 but was independent of the state of lymphatic invasion P=0.169 in the cohort of primary gastric carcinoma. Bioinformatic analysis demonstrated that the target genes of miR-10a are involved in multiple related pathways of tumorigenesis and metastasis. In conclusion our study suggests that miR-10a is involved in the development of gastric cancer and lymph node metastasis particularly in the latter process

Because understanding the pathogenesis of viral-associated hepatocellular carcinomas is important in developing effective means of classification prognosis and therapy we examined the microRNA expression profiles in a large set of 52 human primary liver tumors consisting of premalignant dysplastic liver nodules and hepatocellular carcinomas by quantitative real-time polymerase chain reaction. Initially the accessibility of microRNAs from formalin-fixed paraffin-embedded archival liver tissue by real-time polymerase chain reaction assays was shown. Subsequently target parenchyma from routinely processed tissue was macrodissected RNA was extracted and reverse transcription followed by quantitative real-time polymerase chain reaction was performed. Subsequently five microRNAs miR-122 miR-100 miR-10a miR-198 and miR-145 were selected on the basis of the initial results and further examined in an extended tumor sample set of 43 hepatocellular carcinomas and 9 dysplastic nodules. miR-122 miR-100 and miR-10a were overexpressed whereas miR-198 and miR-145 were up to 5-fold down-regulated in hepatic tumors compared to normal liver parenchyma

Expression levels of three identified miRNAs miR-10a miR-10b and miR-196b were measured in a subset of 37 HCC tumor and non-tumor tissues. Significant downregulation of miR-10a was observed in tumor compared with non-tumor tissues 0.50 vs 1.73 p = 0.031. We further validated our results by RT-qPCR for differentially expressed miRNAs in an independent group of 40 matched fresh frozen tissues 37 plasma samples from NSCLC patients and 28 healthy donors. We found that eight miRNAs miR-21 miR-30d miR-451 miR-10a miR-30e-5p and miR-126* miR-126 miR-145 were differentially expressed by three different statistical analysis approaches. Two of them miR-10a and miR-30e-5p are reported here for the first time. Bead-array results were further verified in an independent group of 40 matched fresh frozen tissues by RT-qPCR. According to RT-qPCR miR-21 was significantly up-regulated P = 0.010 miR-126* P = 0.002 miR-30d P = 0.012 miR-30e-5p P < 0.001 and miR-451 P < 0.001 were down-regulated while miR-10a was not differentiated P = 0.732 in NSCLC tissues. However in NSCLC plasma samples only three of these miRNAs miR-21 miR-10a and miR-30e-5p displayed differential expression when compared to plasma of healthy donors. Moreover we report for the first time that low expression of miR-10a in NSCLC plasma samples was associated with worse DFI P = 0.050 and high expression of miR-30e-5p was found to be associated with shorter OS P = 0.048. In conclusion circulating miR-21 miR-10a and miR-30e-5p in plasma should be further evaluated as potential non-invasive biomarkers in NSCLC

Retinoic acid receptor antagonists inhibit miR 10a expression and block metastatic behavior of pancreatic cancer. Northern blot analysis revealed increased expression levels of miR-10a in metastatic pancreatic adenocarcinoma. The role of miR-10a was analyzed by Morpholino and short interfering RNA transfection of pancreatic carcinoma cell lines and resected specimens of human pancreatic carcinoma. We further show that miR-10a is a retinoid acid target and that retinoic acid receptor antagonists effectively repress miR-10a expression and completely block metastasis. Inhibition of miR-10a expression with retinoic acid receptor antagonists or function with specific inhibitors is a promising starting point for antimetastatic therapies
